} ], "abstractNote": "The Karakoram and the Himalayan mountain range accommodate a large number of glaciers and are the major source of several perennial rivers downstream. To interactively describe to response of glaciers to climate change, a glacier parameterization scheme has been developed and implemented into the regional climate model REMO. The scheme simulates the mass balance as well as changes of the areal extent of glaciers on a subgrid scale. The parameterization scheme is for the first time applied to the region. A regional glacier inventory is compiled and is used to initialize glacier area and volume. Over the highly complex and data sparse region, the simulated mass balance largely agrees with observations including the positive Karakoram anomaly. The simulated equilibrium line altitude is well captured although a systematic underestimation is apparent. Further fractionating a sample based on subcellular compartments can generate molecular information on the state of a tissue and the distribution of its protein components. Although many methods exist for fractionating proteins, the method described here can capture the majority of subcellular fractions simultaneously at reasonable purity. The scalability of this method makes it amenable to small samples, such as embryonic tissues, in addition to larger tissues. The protocol described is for the general fractionation and extraction of proteins from organs or tissues for subsequent analysis by mass spectrometry. It uses differential centrifugation in density gradients to isolate nuclear, cytosolic, mitochondrial and mixed microsomal (Golgi, endoplasmic reticulum, other vesicles and plasma membrane) fractions. Once the fractions are isolated, they are extracted for protein and the samples can then be frozen for processing and analysis at a later date. In mammalian cell nuclei, however, the impact of RNAi has remained more controversial. A key technical hurdle has been a lack of optimized protocols for the isolation and analysis of cell nuclei. Here we describe a simplified protocol for nuclei isolation from cultured cells that incorporates a method for obtaining nucleoplasmic and chromatin fractions and removing cytoplasmic contamination. Cell fractions can then be used to detect the presence and activity of RNAi factors in the nucleus. We include a method for investigating an early step in RNAi, Argonaute protein loading with small RNAs, which is enabled by our improved extract preparations. Since biochemical, cellular and omics studies are usually affected by the lack of suitable fractionation procedures to isolate compartments from these cells, differential and isopycnic centrifugation based techniques were characterized and developed specially for them. Enriched fractions in intact nuclei, mitochondria, peroxisomes,cis-Golgi,trans-Golgi and endoplasmic reticulum (ER) were obtained in differential centrifugation steps and subsequently separated in discontinuous sucrose gradients. Nuclei, mitochondria,cis-Golgi, peroxisomes and smooth ER fractions were obtained as defined bands in 30-60% gradients. Despite the low percentage represented by the microsomes of the total cell homogenate (1.7%), their separation in a novel sucrose gradient (10-60%) showed enough resolution and efficiency to quantitatively separate their components into enriched fractions intrans-Golgi,cis-Golgi and ER. The identity of these organelles belonging to the classical secretion pathway that came from 10-60% gradients was confirmed by proteomics. place for education, fun, and family. Agriculture is the main attraction and remains the constant reason why the Minnesota State Fair is still in existence and remains popular today. However, there is a side to the fair that has not be discussed in rich detail by many authors and that is the existence and social/cultural impact of Indian Villages starting in 1894. Minnesota participated in the 1893 Columbian Exposition geographically located in Chicago, Illinois. The idea came from Indians playing Lacrosse in the Indian Village at the Columbian Exposition and that sparked the discussion and implementation of an Indian Village as an educational experience for the bourgeois fairgoer.
