We determined whether larval Zygoptera responded to the presence of an observer and whether live observation missed some behaviors. We found significant differences between behaviors recorded during live observations and behaviors videotaped in the absence of an observer. All behaviors, except Rotate, were exhibited less frequently in the presence of an observer. These results suggest that larvae respond to the presence of observers as if they were predators. Live observation also missed some behaviors. The duration of Crawl Forward, which can be very subtle, and the frequency of Rotate, which can be very rapid and is easily missed, were greater when recorded from the videotape than by a live observer. Gut and/or faecal analysis can sometimes allow prey remains to be identified visually but is only possible when a component of the diet is resistant to digestion. In some cases there are no solid remains, and when there are it can lead to bias in interpretation of prey choice.2Numerous invasive and non-invasive methods have been developed to characterize predator–prey interactions but two principal areas dominate ‘molecular’ research. These are reviewed under the headings of monoclonal antibodies and DNA-based techniques.3Early ‘molecular’ studies of predator–prey food webs were dominated by the development of monoclonal antibodies. Several studies have documented strong impacts of generalist natural enemies on A. glycines populations using predator exclusion cages and correlating predator and aphid abundances. However, to date no studies directly observed and quantified the natural enemy groups that attack A. glycines in North America under field conditions. In 2005, we conducted 72 h of direct observations of predation on natural populations of A. glycines in Michigan. The observations were conducted during three consecutive weeks during morning, afternoon and evening, in replicated 1 m2 areas. A total of 643 predators within 12 groups and 211 predation events on A. glycines were observed. Transient predators such as Harmonia axyridis and Coccinella septempunctata (Coleoptera: Coccinellidae) accounted for most of the observed mortality and were very effective on a per capita basis, despite relatively short residence time in the observed patches. Transient predators responded positively to increased A. glycines field densities. Resident predators, particularly Orius insidiosus (Hemiptera: Anthocoridae), were abundant and accounted for many predation events, but they were not as effective on a per capita basis and did not respond to changes in aphid density. Lower predation was obtained on A. glycines located at the bottom portion of the plants and on alate aphids. In practice, however, inter- and intraspecific interactions between generalist predators, and between the predators and their prey, within multispecies systems under the influence of rapidly changing biotic and abiotic variables are difficult to predict. We discuss trade-offs between the relative merits of specialists and generalists that allow both to be effective, and often complementary, under different circumstances. A review of manipulative field studies showed that in ∼75% of cases, generalist predators, whether single species or species assemblages, reduced pest numbers significantly. Techniques for manipulating predator numbers to enhance pest control at different scales are discussed. Given that several investigations have determined that cells sensitive to pheromone can resolve only low rates of encounter with pheromone filaments, generally up to 10 pulses/s, it would appear that individual filaments encountered at higher rates would not be resolved by the insects' sensory system and therefore the stimulus would be perceived as a non-flickering signal. Behaviourally, this has been thought to be expressed as the cessation of upwind progress. Previous studies with Cadra cautella (Walker) (Lepidoptera: Pyralidae) demonstrated that upwind flight by these males is not inhibited in rapidly pulsed plumes. Therefore, to determine whether a flickering signal is required for upwind progress by C. cautella, males were introduced to homogeneous clouds of pheromone in a wind tunnel and their behaviour recorded. For comparison, Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae), a species from a long-diverged lineage, was also used. Upwind progress by C. cautella is not impeded by the constant olfactory signal provided by a homogeneous cloud of pheromone, but this is not true for P. gossypiella. Furthermore, although C. cautella directs its flight upwind in a homogeneous cloud, its heading is not always due upwind. Potential mechanisms are discussed. A large proportion of each beetle population tested positive, suggesting that collops and ladybird beetles are active predators on both B. tabaci and P. gossypiella. Technical innovations were performed in image analysis and in the use of robotics. In image analysis, both the automatic detection of the insect and the calculation of several variables (linear and angular speed, locomotor activity, etc.) are carried out in real-time, which saves time and allows long-term studies without any intervention of the operator. The displacement of the camera by robotics allows the simultaneous study of many individuals (i.e. sixty individuals for the study of locomotor activity of adult insects, more than 400 individuals for the study of the emergence of adult parasitoids from their host). Four applications of this new device are presented. They deal with circadian rhythms in parasitoid insects: locomotor activity and time variation in locomotory parameters, daily distribution of the emergence and onset of the locomotor activity in adults. It has remained challenging, however, to acquire data from small and fast-moving animals, such as insects in flight, due to the limited spatial and temporal resolution of the systems currently available. Our research on free-flying insects motivated the development of new methods in the context of two different experimental settings. First, the position and precise body axis direction of honey bees approaching a food source were automatically measured. Second, the flight trajectories of a phonotactic parasitoid fly homing in on its cricket host were recorded in 3D. We used pan-tilt cameras, i.e. cameras with moveable optics, to follow the animal's path with a close up image. Novel methods were developed for image acquisition and position measurement using pan-tilt cameras, as well as calibration and data evaluation in 3D world coordinates. The innovations of this system comprise: (1) Acquisition of images in high spatial detail over large observation areas. (2) Image acquisition at a field rate of 50 Hz PAL. (3) Free positioning of the cameras for 3D acquisition. (4) Computation of the flight path in 3D world coordinates. We illustrate the capabilities of the system with data obtained from a calibration object as well as from the behaviour of unrestricted, free-flying flies and bees.
