Oxygen equilibrium curves were measured by Imai's continuous recording method (Imai, K. (1981) Methods Enzymol. 76, 438-449). The rate of the R to T transition was determined after full laser photolysis of the carbon monoxide derivative by measuring the resultant absorbance changes at an isosbestic point for ligand binding. Chemical cross-linking caused the R-state O2 affinity of alpha subunits to decrease 6-fold compared with unmodified hemoglobin. This inhibition of O2 binding was the result of both a decrease in the rate constant for ligand association and an increase in the rate constant for dissociation. The O2 affinity of R-state beta subunits was reduced 2-fold because of an increase in the O2 dissociation rate constant. These changes were attributed to proximal effects on the R-state hemes as the result of the covalent cross-link between alpha chain G helices. This proximal strain in cross-linked hemoglobin was also expressed as a 5-fold higher rate for the unliganded R to T allosteric transition. The fourth O2 equilibrium binding constant, K4, measured by kinetic techniques, could be used to analyze equilibrium curves for either native or cross-linked hemoglobin. The resultant fitted values of the Adair constants, a1, a2, and a3 were similar to those obtained when K4 was allowed to vary, and the fits were of equal quality. In this situation, lateral diffusion equilibration is relatively rapid and the distribution of exchanging materials is governed chiefly by the permeability of the capillary walls. We modeled extravascular distribution of exchanging substances from this kind of capillary and illustrated the changes expected in the outflow profile with increasing permeability, the evolution from the barrier-limited to the flow-limited case. We then examined the two extremes of the assemblies of such capillaries in an organ. In one, the large-vessel transit times are constant and the capillary transit times account for the outflow distribution of the vascular reference substance; in the other, the capillary transit times are constant but the large-vessel transit times vary. The barrier-limited and flow-limited cases corresponding to these are very different. In the case intermediate between these two extremes, the transit times in both the large vessels and the capillaries in the organs vary. If the organ is functionally homogeneous, the distribution of capillaries supplied by each large vessel is the same, and the situation may be described by a product distribution. The cross-sectional area and capacity in the various portions of the vascular bed were calculated. The greatest total cross-sectional area in the entire vascular bed was found in the venules, a result not in agreement with similar reports made by others using measurements from vessels in fixed preparations. This discrepancy probably results from disagreement regarding the diameter of capillary vessels. If correct, Pic can only contribute to lowering pleural pressure (Ppl), to the extent that it lowers abdominal pressure (Pab). To test this we measured Pab and Ppl during during Mueller maneuvers in which deltaPab = 0. Because there was no outward displacement of the rib cage, Pic must have contributed to deltaPpl, as did Pdi. Under these conditions the total pressure developed by the inspiratory muscles across the rib-cage pathway was less than Pdi + Pic. Therefore, we rejected the hypothesis. A plot of Pab vs. Ppl during relaxation allows partitioning of the diaphragmatic and intercostal/accessory muscle contributions to inspiratory pressure swings. The analysis indicates that the diaphragm can act both as a fixator, preventing transmission of Ppl to the abdomen and as an agonist.
