manganese porphyrins,\ndiffering in their meso substituents, with peroxynitrite\n(ONOO–) and carbonate radical anion\n()\nusing stopped-flow and pulse radiolysis, respectively. Rate constants for the\nreactions of Mn(III) porphyrins with ONOO– ranged from 1\n× 105 to 3.4 × 107\nm–1 s–1 and correlated\nwell with previously reported kinetic and thermodynamic data that reflect the\nresonance and inductive effects of the substituents on the porphyrin ring.\nRate constants for the reactions of Mn(III) porphyrins with\n\nranged from 2 × 108 to 1.2 × 109\nm–1s–1 at pH ≤ 8.5\nand increased with pH as a consequence of the ionization of the complexes.\nMn(II) porphyrins reacted with\n\nwith rate constants ranging from 1 × 109 to 5 ×\n109 m–1s–1\nat pH 10.4. Hence, fast scavenging of ONOO– and\n\nby manganese porphyrins could occur in vivo because of the catalytic\nreduction at the expense of a number of cellular reductants. Additionally, we\ndetermined the pKa of the axial water molecules\nof the Mn(III) complexes at pH 7.5–13.2 by spectrophotometric titration.\nResults were consistent with two acid-base equilibria for most of the\ncomplexes studied. The pKa values also correlated\nwith the resonance and inductive effects of the substituents. For reasons unknown, only some proteins are prone to carbonylation.\n\n\n\nWe used mass spectrometry analysis to identify carbonylated sites in: BSA that had undergone in vitro MCO, and 23 carbonylated proteins in Escherichia coli. The presence of a carbonylated site rendered the neighbouring carbonylatable site more prone to carbonylation. Most carbonylated sites were present within hot spots of carbonylation. These observations led us to suggest rules for identifying sites more prone to carbonylation. We used these rules to design an in silico model (available at http://www.lcb.cnrs-mrs.fr/CSPD/), allowing an effective and accurate prediction of sites and of proteins more prone to carbonylation in the E. coli proteome.\n\n\n\nWe observed that proteins evolve to either selectively maintain or lose predicted hot spots of carbonylation depending on their biological function. Stopped-flow kinetic studies show that the reaction between peroxynitrite and GSH-Px is first-order in each of the reactants, with an apparent second-order rate constant of 4.5 +/- 0.2 x 10(4) M-1 s-1 per monomer unit of enzyme. In good agreement with this value, GSH-Px inactivation experiments afford an apparent second-order rate constant of 1.8 +/- 0.1 x 10(4) M-1 s-1 per monomer unit of enzyme. The hydroxyl radical scavengers mannitol, DMSO, and benzoate (at 100 mM) afford only 8-12% protection of the enzyme, while addition of 25 mM bicarbonate results in 55% protection. The minimal protection by hydroxyl radical scavengers indicates, as expected, that hydroxyl radicals are not involved in the inactivation. Protection by bicarbonate occurs because peroxynitrite is rapidly trapped by CO2 to form the adduct nitrosoperoxycarbonate (ONOOCO2-), and/or other reactive species that preferentially decompose to nitrate rather than react with GSH-Px. The close agreement between the rate constants obtained from enzyme inactivation and from stopped-flow kinetics experiments suggests that the mechanism of the reaction between peroxynitrite and GSH-Px involves the oxidation of the ionized selenol of the selenocysteine residue in the enzyme's active site (E-Se-) by peroxynitrite. This reaction does not simply involve formation of the selenenic acid, E-SeOH, because E-SeOH is an intermediate in the catalytic cycle of the enzyme, and thus its formation cannot explain the inactivation we observe.
