"abstractNote": "Human umbilical vein endothelial cell (HUVEC)-based gene expression studies carried out under hypoxia and/ or hyperglycemia bear huge potential in modelling endothelial cell response in cardiovascular disease and diabetes. However, such studies require reference genes that are stable across the whole range of experimental conditions. These reference genes have not been comprehensively defined to date. We applied human genome-wide microarrays and quantitative real-time PCR (qRT-PCR) on RNA obtained from primary HUVEC cultures that were incubated for 24 h either in euglycemic or hyperglycemic conditions and then subjected to short-term CoCl2-induced hypoxia of either 1, 3 or 12 h. Using whole-transcript arrays, we selected ten commonly used reference genes with no significant expression variation across 8 different conditions. These genes were ranked using NormFinder software according to their stability values. Consequently, five genes were selected for validation by quantitative real-time PCR (qRT-PCR). These were: ribosomal protein large P0 (RPLP0), transferrin receptor (TFRC), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), β-glucuronidase (GUSB), and β-actin (ACTB). All five genes displayed stable expression under hyperglycemia. However, only RPLP0 and TFRC genes were stable under hypoxia up to 12 h. Under hyperglycemia combined with hypoxia up to 12 hours the expression of RPLP0, TFRC, GUSB and ACTB genes remained unchanged. Our findings strongly confirm that RPLP0 and TFRC are the most suitable reference genes for HUVEC gene expression experiments subjected to hypoxia and/or hyperglycemia for the given experimental conditions. We provide further evidence that even commonly known references genes require experimental validation for all conditions involved.",

"abstractNote": "BACKGROUND: Cardiovascular disease and type 2 diabetes are examples of chronic diseases that impose significant morbidity and mortality in the general population worldwide. Most chronic diseases are associated with underlying preventable risk factors, such as elevated blood pressure, high blood glucose or glucose intolerance, high lipid levels, physical inactivity, excessive sedentary behaviours, and overweight/obesity. The occurrence of intermediate outcomes during childhood increases the risk of disease in adulthood. Sugar-sweetened beverages are known to be significant sources of additional caloric intake, and given recent attention to their contribution in the development of chronic diseases, a systematic review is warranted. We will assess whether the consumption of sugar-sweetened beverages in children is associated with adverse health outcomes and what the potential moderating factors are.Methods/design: Of interest are studies addressing sugar-sweetened beverage consumption, taking a broad perspective. Both direct consumption studies as well as those evaluating interventions that influence consumption (e.g. school policy, educational) will be relevant. Non-specific or multi-faceted behavioural, educational, or policy interventions may also be included subject to the level of evidence that exists for the other interventions/exposures. Comparisons of interest and endpoints of interest are pre-specified. We will include randomized controlled trials, controlled clinical trials, interrupted time series studies, controlled before-after studies, prospective and retrospective comparative cohort studies, case-control studies, and nested case-control designs. The MEDLINE(R), Embase, The Cochrane Library, CINAHL, ERIC, and PsycINFO(R) databases and grey literature sources will be searched. The processes for selecting studies, abstracting data, and resolving conflicts are described. We will assess risk of bias using design-specific tools. To determine sets of confounding variables that should be adjusted for, we have developed causal directed acyclic graphs and will use those to inform our risk of bias assessments. Meta-analysis will be conducted where appropriate; parameters for exploring statistical heterogeneity and effect modifiers are pre-specified. The Grading of Recommendations Assessment, Development and Evaluation (GRADE) approach will be used to determine the quality of evidence for outcomes.Systematic review registration: PROSPERO CRD42014009641.",

"abstractNote": "PURPOSE: To examine the benefits of sexual activity on glycated hemoglobin (HbA1c)in penile prosthesis implanted patients with type 2 diabetes mellitus (DM).\nMATERIALS AND METHODS: Sixty-seven male subjects who had HbA1c levels of ≥ 6.5% before and could perform regular sexual activity after the implantations were enrolled. The contribution of sexual activity on glycemic control assessed by HbA1clevel as well as age, duration of DM and frequency of sexual activity were evaluated.\nRESULTS: Mean age and mean time from the surgery of the study patients was 59.9 years (range,30-82) and 22.6 months (range, 10-63), respectively. The average of penile prosthesis usage for sexual activity was 9.9 times per month (range, 2-28). Compared with the preimplantation, the absolute mean change in HbA1c after penile prosthesis implantation was found as - 0.2% (P > .05). This study also revealed that more sexual activity was associated with more reduction in HbA1c.\nCONCLUSION: The present study demonstrated that sexual activity is associated with HbA1c reduction, which is clinically important in patients with type 2 DM after penile prosthesis implantation. ",

"abstractNote": "PURPOSE: To evaluate the impact of metabolic syndrome (MS) on erectile dysfunction (ED) and lower urinary tract symptoms (LUTS).\nMATERIALS AND METHODS: We included patients who had presented at the urology outpatients with LUTS or ED complaints and at the endocrinology outpatients for diabetes between May 2012 and April 2013. MS was present in 50 of the 107 patients (42.7%). The blood pressure, fasting blood sugar, serum lipid profile, triglyceride, total cholesterol, body mass index (BMI) and total prostate specific antigen (PSA) values were recorded. The international prostate symptom score (IPSS), quality of life score and international erectile function index (IIEF-5) values were determined for the patients. All patients also underwent uroflowmetry together with prostate volume and residual urine volume measurement.\nRESULTS: There was a significant negative correlation between the IPSS and IIEF scores of the patients (P < .001, r = -0.42). There was no significant difference regarding IPSS scores between patients with and without MS (P = .6), while the IIEF-5 scores were significantly lower in the MS group (P = .03).\nCONCLUSION: We found that metabolic syndrome did not significantly affect LUTS but could significantly contribute to ED. We therefore feel patients presenting with ED complaints should also be carefully evaluated for MS. ",
