In Europe, this applies above all to species of the genus Trichobilharzia. For Austria, a lot of data are available on cercarial dermatitis and on the occurrence of Trichobilharzia, yet species identification of trematodes in most cases is doubtful due to the challenging morphological determination of cercariae. During a survey of trematodes in freshwater snails, we were able to detect a species in the snail Physella acuta (Draparnaud, 1805) hitherto unknown for Austria, Trichobilharzia physellae; this is also the first time this species has been reported in Europe. Species identification was performed by integrative taxonomy combining morphological investigations with molecular genetic analyses. The results show a very close relationship between the parasite found in Austria and North American specimens (similarity found in CO1 ≥99.57%). It was our goal to confirm the tentative diagnosis of parafilariosis by identifying Parafilaria bovicola in exudate samples using molecular methods for the first time in Austria. We asked veterinarians in the field to collect exudate from typical lesions on cattle. We performed polymerase chain reactions (PCRs) and sequenced a 674-bp section of the mitochondrial cytochrome oxidase subunit I in all positive samples. Overall, in 57 of 86 samples, P. bovicola was confirmed by PCR in cattle from Lower Austria, Upper Austria, Styria, Salzburg, Carinthia, and Tyrol. Sequencing detected four different haplotypes or genotypes, respectively, indicating multiple routes of introduction. Therefore, this work aimed to close that gap by recording and analysing the parasite community of a large number of birds from Austria for the first time. (31.7%) and Fimbriarioides intermedia (1.7%); Acanthocephala: Filicollis anatis (5%), Polymorphus minutus (30%) and one cystacanth unidentified (1.7%); Trematoda: Apatemon gracilis (3.3%), Echinostoma grandis (6.7%), Echinostoma revolutum (6.7%) and Notocotylus attenuatus (23.3%); Nematoda: Porrocaecum crassum (1.7%) and one not identified (1.7%). The frequency distribution of parasites showed a typical pattern in which 39 birds (65%) were either not parasitized or were harbouring up to five worms, whereas more intense infestations occurred in a lesser number of hosts. In Europe, there are 263 valid named species recognized, many of which are difficult to identify using morphology only. Here we explore the use of DNA barcodes as a tool for identification and species discovery in European gracillariids. We present a barcode library including 6791 COI sequences representing 242 of the 263 (92%) resident species. Our results indicate high congruence between morphology and barcodes with 91.3% (221/242) of European species forming monophyletic clades that can be identified accurately using barcodes alone. The remaining 8.7% represent cases of non-monophyly making their identification uncertain using barcodes. Species discrimination based on the Barcode Index Number system (BIN) was successful for 93% of species with 7% of species sharing BINs. We discovered as many as 21 undescribed candidate species, of which six were confirmed from an integrative approach; the other 15 require additional material and study to confirm preliminary evidence. Most of these new candidate species are found in mountainous regions of Mediterranean countries, the South-Eastern Alps and the Balkans, with nine candidate species found only on islands. In addition, 13 species were classified as deep conspecific lineages, comprising a total of 27 BINs with no intraspecific morphological differences found, and no known ecological differentiation. Double-digest restriction-site associated DNA sequencing (ddRAD) analysis showed strong mitonuclear discrepancy in four out of five species studied. Compared to classical methodologies based on manual sorting and morphological identification of benthic taxa, eDNA metabarcoding offers potentially important advantages for assessing the environmental quality of sediments. The methods and protocols utilized for sediment eDNA metabarcoding can vary considerably among studies, and standardization efforts are needed to improve their robustness, comparability and use within regulatory frameworks. Here, we review the available information on eDNA metabarcoding applied to sediment samples, with a focus on sampling, preservation, and DNA extraction steps. We discuss challenges specific to sediment eDNA analysis, including the variety of different sources and states of eDNA and its persistence in the sediment. It has been repeatedly observed that more than one BIN can be found under the same species name particularly when large geographic scales are considered. One such case concerns Eupithecia conterminata, a species widespread in North European countries and restricted to mountainous regions in the rest of the continent, for which five BINs are found in Europe. In order to solve the question concerning the taxonomic status of these BINs and European populations, we employed an integrated approach by combining classical morphological traits (genitalia and wing markings) with those of molecular data, the latter involving both mitochondrial and nuclear genes. This approach allowed us to recognize two valid species in Europe, E. conterminata, currently known only in Fennoscandia, Baltic countries and Russia, and Eupithecia manniaria sp. rev., with distribution covering Central and South European countries. We furthermore synonymized Eupithecia pindosata syn. nov. from Greece with E. manniaria.
