A C18 column was used with an acetonitrilewater gradient mobile phase. Paeoniflorin and albiflorin were quantified at 230 nm. Ions with m/z 121 and 327 were produced with the MS detector, using the paeoniflorin precursor ion with m/z 479. For albiflorin, the precursor ion with m/z 479 produced the m/z 121 and 77 ions. The amounts of paeoniflorin and albiflorin found in the raw herb were 33.2 and 1.8 mg/g, respectively; and in the dried aqueous extract, the amounts were 34.8 and 15.7 mg/g, respectively. However, root cortices of P. delavayi and P. decomposita also are used under the name of this drug in some regions such as Yunnan and Sichuan Provinces, respectively. In order to make a comparison of their chemical constituents, the compounds of the three Paeonia species were analyzed by high-performance liquid chromatography-diode array detection/electrospray ionization and quadrupole-time-of-flight tandem mass spectrometry (HPLC-DAD/ESI-MS2). A total of 50 compounds were observed in the 50% (v/v) methanolic extracts, including 17 monoterpenes, 14 galloyl glucoses, 10 acetophenones, 5 phenolic acids, 3 flavonoids and 1 triterpene. These chemical constituents were separated on a C18 column and identified or tentatively characterized based on UV spectra and MS fragmentation behavior. The chemical compositions of the three Paeonia species were found to have many differences. Paeonol was the predominant constituent of P. suffruticosa and P. decomposita, while P. delavayi contained albiflorin and more galloyl glucoses than the other two Paeonia species. Most of these identified compounds have been reported from P. delavayi and P. decomposita for the first time. The ESI-MS fragmentation behavior of monoterpene glycosides, acetophenones and galloyl glucoses was also investigated successively, and appropriate characteristic pathways were proposed. Paeonol relaxed isolated rat aorta rings by 95.6% while the 10(-6) M forskolin-induced vasodilatation used as 100%. The EC(50) of vasodilatation by paeonol is 2.9x10(-4) M. Although paeonol exerted endothelium-independent relaxation, L-NAME treatment inhibited paeonol-induced vasodilation of endothelium intact rings, while indomethacin did not. Both L-NAME and ODQ did not affect paeonol relaxation in the rings without endothelium. In addition, paeonol markedly elevated NO generation in cultured endothelial cells. Pre-treatment of propranolol, glibenclamide, TEA and BaCl(2) did not affect paeonol relaxation of endothelium removed rings. On the other hand, pre-treated of rings (without endothelium) with paeonol markedly blocked vasoconstriction induced by AngII, PGF(2alpha), 5-HT, dopamine, vasopressin, endothelin-1 and PE. The paeonol incubation also significantly attenuated KCl-induced contraction which mainly depended on Ca(2+) influx. In Ca(2+)-free medium (containing 10(-4) M of EGTA and 60 mM of KCl), paeonol suppressed the contraction curve of CaCl(2). In addition, paeonol also inhibited contraction by PE in Ca(2+) free solution (containing 10(-4) M of EGTA) which mainly relied on intracellular Ca(2+) release. Whole-cell patch-clamp experiment showed that paeonol shifted the I-V curve and the peak value of calcium currents was significantly inhibited. "Chronic cerebral hypoperfusion, a mild ischemic condition, is associated with the cognitive deficits of AD. Paeoniflorin (PF), a major constituent of peony root, was proved to be neuroprotective in middle cerebral artery occlusion model. In this study, we investigated whether PF could attenuate chronic cerebral hypoperfusion-induced learning dysfunction and brain damage in rat. Seven weeks after permanent bilateral occlusion of the common carotid arteries, the rats were tested in the Morris water maze. Subsequently, the animals were sacrificed and neurons, astrocytes and microglias were labeled with immunocytochemistry in hippocampus. PF at the dose of 2.5 mg/kg ameliorated cerebral hypoperfusion-related learning dysfunction and prevented CA1 neuron damage. Chronic cerebral hypoperfusion increased the immunoreactivity of astrocytes and microglias in hippocampus. The increase was prevented by PF at the dose of 2.5 mg/kg. Cerebral hypoperfusion also increased expression of nuclear factor-[kappa]B (NF-[kappa]B), mostly in astrocytes, but not in neurons. With the treatment of PF (2.5 mg/kg), NF-[kappa]B immunostaining was diminished in hippocampus. Two bionic technologies titled 'in vitro digestion' and 'extraction with biomembrane' were developed for pre-treatment of herbal decoction. The decoctions of Aconiteum carmichaeli and Paeonia lactiflora were digested at body temperature, at the acidity of the stomach or intestine and with inorganic and organic materials (digestive enzymes were included for whole-bionic and excluded for semi-bionic) found in the stomach or intestine. Being similar to the biomembrane between the gastrointestinal tract and blood vessels, monolayer liposome was used as a biomembrane model. Affinity-monolayer liposome metals and water-soluble metals were used for speciation analysis and bioavailability assessment of copper and zinc in herbal decoction. In the decoction of Aconiteum carmichaeli and Paeonia lactiflora, Zn was mainly absorbed in the intestine and Cu was mainly absorbed by both stomach and intestine. However, the neuronal death is prevented when peony root extract (PR) is administered prior to cobalt application. We tested the hypothesis that PR modulates the expression of neuroprotective proteins in the cerebrum of mouse cobalt focus by proteomic analysis using two-dimensional polyacrylamide gel electrophoresis and mass spectrometry to screen for differentially expressed proteins. Analyses revealed that transthyretin, a carrier protein for thyroid hormones and retinoids, and the brain form of phosphoglycerate mutase, a glycolytic enzyme, were upregulated in the cobalt-treated mouse cerebrum and further increased by PR administration in association with upregulation of neurogranin/RC3, a target of the transcriptional activation by thyroid hormones and retinoids.
